Timolol maleate is a B1 & B2 (non-selective) adrenergic receptor blocking agent that doesn't have a significant intrinsic sympathomimetic or local anaesthetic activity. Therefore, reducing aqueous humour formation.

Dorzolamide is a potent inhibitor of human carbonic anhydrase enzyme (CA-II) in the ciliary processes, that leads to decrease in aqueous humour secretion by slowing the formation of bicarbonate ions with subsequent reduction in sodium and fluid transport. This reduces aqueous humor formation which results in a reduction of intraocular pressure.
